Genetic diversity of Justicia wrightii (Acanthaceae), an endemic of the southern Pecos basin of Texas and New Mexico. Prior to 2000 Justicia wrightii Gray was known from only nine vouchered specimens, eight in Texas, the most recent in 1953, and one in New Mexico in 1957. Recent surveys found the population in New Mexico to be restricted to a roughly 50 km x 25 km area with the greatest concentration of plants near Carlsbad, New Mexico. In 2003, seven of the eight previously vouchered locations in Texas were searched. No plants were found, but the species was found at two new locations in Texas. The objective of this study was to sample the amount and distribution of genetic differentiation within and among populations of J. wrightii. Forty individuals from each of the Texas locations and forty individuals from the densest concentration of plants in New Mexico were sampled. In addition, eleven individuals were sampled from each of five outlying locations in New Mexico. Three inter-simple sequence repeat primers were used to assess genetic differentiation. There was no genetic differentiation within and among Texas locations and within and among the New Mexico locations. However, genetic differentiation between Texas and New Mexico locations was detected. These results indicate a genetic break between Texas and New Mexico populations and suggest that populations, if they exist, between the Texas and New Mexico locations are isolated and unable to move genetic material across the 100 km separating the known populations. Lastly, these results have implications concerning the protective status of J. wrightii.
