Tragopogon dubius is a diploid biennial that was introduced to the U.S. from Eurasia and has served as the common parent of the allotetraploid species T. mirus and T. miscellus, both of which formed repeatedly during the last 80 years in the Pacific Northwest region of the U.S. To examine genetic variation within and between populations of T. dubius, we created a microsatellite-enriched library, from which variable markers were developed. We used these microsatellite markers to analyze genetic differences within and between Old World and New World populations of T. dubius. These markers will also be used in the construction of genetic linkage maps for T. dubius and will contribute to ongoing work to improve our understanding of recent and recurrent polyploidy in Tragopogon. We found that one population of T. dubius, which was collected in Spain and exhibits morphological differences from known T. dubius, also displays molecular differentiation from known T. dubius.
