Samad, Abdul [1]. A Protocol for Nonradioactive Differential Display Optimized for Cotton (Gossypium hirsutum L.) Fiber RNA. A protocol for nonradioactive differential display visualized by ethidium bromide staining was optimized for cotton (Gossypium hirsutum L.) fiber total RNA. The procedure was used to identify genes differentially expressed at 0 DAP (Day After Pollination) and 10 DAP of developing cotton fiber. Different populations of total RNA were reversely transcribed using three single anchored oligo-dT primers. PCR amplification of relative cDNA was carried out in combination with five arbitrary primers (random decamers). New one percent agarose containing ethidium bromide for staining was a simplified protocol that can resolve amplified transcripts over a large range of molecular weights.
